There are many options for travelling to Rome from Venice. Although i love driving, but that might be a HUGE concern in Rome.

Very pleasant journey and make sure you take it from the Santa Lucia Station in Venice. The other nearest station is in Maestre and it is on the mainland. This train goes all the way into the heart of Rome i.e. the Rome Termini Train Station.
